Geometry Adaptation of Protrusion and Polarity Dynamics in Confined Cell Migration

Cell migration in confining physiological environments relies on the concerted dynamics of several cellular components, including protrusions, adhesions with environment, and cell nucleus. However, it remains poorly understood how dynamic interplay these components polarity determine emergent behavior at scale. Here, we combine data-driven inference a mechanistic bottom-up approach to develop model for protrusion confined migration, revealing adapt geometries. Specifically, use experimental data joint protrusion-nucleus trajectories cells micropatterns systematically linking stochastic polarity, This indicates that constrictions through switch from negative positive self-reinforcing feedback loop. Our further reveals this loop leads stereotypical cycles drive constrictions.
